Effect of the Safener Dichlormid on Maize Peroxygenase and Lipoxygenase*

چکیده

S-Ethyl-N,N-dipropylthiocarbamate (EPTC) was oxidized into its corresponding sulfoxide by microsomal fractions from etiolated maize seedlings. This reaction is catalyzed by a hydroperoxide-dependent enzyme, identified as a peroxygenase. The hydroperoxides formed from fatty acids by a lipoxygenase are efficient co-substrates o f the EPTC sulfoxidation. The effects o f the safener dichlormid on the peroxygenase and lipoxygenase activities were studied in vitro and in vivo. In vitro, the safener is not an inhibitor o f these enzymes. Dichlormid seems to act, in vivo, by modulating the amounts o f peroxygenase and lipoxygenase in treated plants.

The inhibition of fatty acid elongation by pebulate can be effectively counteracted by the safener dichlormid.

The thiocarbamate herbicide pebulate inhibits fatty acid elongation, which is necessary for surface lipid biosynthesis. As both barley and wild oats are susceptible to pebulate, the safener dichlormid was used to study the reversal of its herbicidal effect. Fatty acid elongation was restored by a dichlormid pretreatment in barley, but not in pebulate-expressed oats.

Co-Induction of a Glutathione-S-transferase, a Glutathione Transporter and an ABC Transporter in Maize by Xenobiotics

Glutathione conjugation reactions are one of the principal mechanisms that plants utilize to detoxify xenobiotics. The induction by four herbicides (2,4-D, atrazine, metolachlor and primisulfuron) and a herbicide safener (dichlormid) on the expression of three genes, ZmGST27, ZmGT1 and ZmMRP1, encoding respectively a glutathione-S-transferase, a glutathione transporter and an ATP-binding casset...

Milestones in the Development of Herbicide Safeners*

The concept o f using chemical safeners to improve herbicide selectivity became widely known in 1969 with the introduction o f naphthalic anhydride (N A ) to improve the tolerance o f maize to thiocarbamate herbicides. Soon thereafter dichlormid was also developed as a safener for thiocarbamates in maize. At present there are at least eight different chemical safeners that have been developed c...

Herbicide safener-binding protein of maize. Purification, cloning, and expression of an encoding cDNA.

Dichloroacetamide safeners protect maize (Zea mays L.) against injury from chloroacetanilide and thiocarbamate herbicides. Etiolated maize seedlings have a high-affinity cytosolic-binding site for the safener [3H](R,S)-3-dichloroacetyl-2,2,5-trimethyl-1, 3-oxazol-idine ([3H]Saf), and this safener-binding activity (SafBA) is competitively inhibited by the herbicides. The safener-binding protein ...

The effects o f individual or combined treatment o f the cyclohexanedione herbicide sethoxy dim and the safener dichlormid on total lipid synthesis, protein synthesis and acetyl-CoA car boxylase (ACCase, EC 6.4.1.12) activity o f grain sorghum [Sorghum bicolor (L.) M oench, var. G623] were investigated. Sethoxydim and dichlormid were tested at concentrations o f 0, 5, 50, and 100 |iM each. Se...
